When should I take PT-141 and how often is too often?

Updated 2026-05-05

Take PT-141 at least 45 minutes before sexual activity. The Vyleesi prescribing label allows up to one dose every 24 hours and no more than 8 doses per month. Most people in the trials used it 2-3 times per month, never more than once a week. Dosing more often increases your risk of skin darkening (focal hyperpigmentation) and longer stretches of raised blood pressure. PT-141 is on-demand, not a daily peptide.

IfIf you want to use PT-141 tonight
Thenthen inject 1.75 mg subcutaneously at least 45 minutes before activity
IfIf you already dosed within the last 24 hours
Thenthen do not stack another dose - the safety data does not support it
IfIf you find yourself wanting it more than 8 times a month
Thenthen talk to a doctor about underlying causes (libido, hormones, stress) rather than upping the frequency
IfIf the effect feels weak
Thenthen check timing first (was it really 45+ minutes?) before increasing dose
Key facts
  • Vyleesi label: 1.75 mg subcutaneous, max 1 dose/24 hours, max 8 doses/month
  • Median use in the phase 3 trials was 10 injections over 24 weeks - lower than many people expect
  • Optimal timing window has not been fully characterized; 45 min is the labeled minimum
  • Effect duration is variable; patients are encouraged to learn their own response curve
  • Compounded versions follow the same on-demand pattern, even if the dose is different
